Clinical and pharmacological groups / Group affiliation
Serotonin 5-HT1 receptor agonist. Drug with anti-migraine activity
Active ingredient
sumatriptan
Pharmacotherapeutic group Amigrenin
Antimigrainous agent
Pharmacological effect Amigrenin
Migraine treatment agent; serotonin 5-HT1 receptor agonist. The therapeutic effect of sumatriptan appears to be due to two mechanisms: it triggers serotonin 5-HT1 receptors in the smooth muscle of the carotid vascular system and causes them to narrow. Carotid arteries carry blood to tissues of the head, including the meninges; migraine attacks have been linked to meningeal vasodilation. In addition, sumatriptan activates afferent trigeminal nerve terminal receptors in the dura matrices, resulting in decreased release of sensory neuropeptides. Sumatriptan does not penetrate well through the BSE and has no analgesic properties.
Sumatriptan is usually effective 30 minutes after ingestion and 10 to 15 minutes after administration.
Testimony sumatriptan
Treating migraine attacks and Horton syndrome (histamine cephalgia).
Method of use, course and dosage sumatriptan
The dose taken by mouth is 100 mg once for a migraine attack; sometimes less than 100 mg may be effective; if an attack recurs, it may be repeated, up to a maximum of 300 mg daily. A dose of six mg of n/a is given; if necessary, the same dose may be repeated no earlier than one hour after the first dose; maximum daily n/a dose is 12 mg.
For Horton syndrome, a dose of 6 mg once during an attack is given; maximum daily dose is 12 mg; interval between injections is at least one hour.
