Pharmacodynamics
Mechanism of Action
Valacyclovir is an anti-virus drug that is an acyclovir ester with L-valine. Acyclovir is an analog of guanine, a purine nucleoside.
In humans, valacyclovir rapidly converts to acyclovir and valine, probably under the influence of the enzyme valacyclovirhydrolase. Acyclovir is a specific herpesvirus inhibitor, with activity against HSV-1, HSV-2, VZV, CMV, EBV, and HHV-6. It prevents viral DNA synthesis immediately after phosphorylation and conversion to an active form, acyclovirtriphosphate. The first stage of phosphorylation requires the activity of virus-specific enzymes. For example, for HSV-1, HSV-2, and EBV, the viral thymidine kinase enzyme is present only in infected cells by the virus. The partial selectivity of phosphorylation is carried out in CMV indirectly through the phosphotransferase gene product UL97. The need for acyclovir to be activated by a specific viral enzyme largely explains its selective action.
