Indications for use Tamsulosin:

Pharmacokinetics:

Absorption - high (over 90%). TCmax after a single oral administration of 0.4 mg - 6 hours. The connection with plasma proteins is 94-99%. The volume of distribution is insignificant - 0.2 l / kg. Slowly metabolized in the liver with the formation of pharmacologically active metabolites. Most of it is present in the blood unchanged. It is excreted by the kidneys (4-9% unchanged). T1 / 2 - 10-12 hours for extended-release capsules and 15-19 hours for controlled release tablets.

Pharmacodynamics:

Selectively and competitively blocks postsynaptic alpha1a-adrenergic receptors located in the smooth muscles of the prostate gland, the bladder neck and the prostatic urethra. Reduces the tone of the smooth muscles of the prostate gland, bladder neck and prostatic urethra, improving urine flow. At the same time, the symptoms of obstruction and irritation associated with benign prostatic hyperplasia are reduced. The therapeutic effect occurs 2 weeks after the start of treatment. Tropism for alpha1a-adrenergic receptors located in the bladder is 20 times greater than its ability to interact with alpha1b-adrenergic receptors located in vascular smooth muscles.

Contraindications:

Hypersensitivity.

Side effects Tamsulosin:

Headache, dizziness, asthenia, sleep disturbances (drowsiness or insomnia), retrograde ejaculation, decreased libido, priapism, back pain, rhinitis; orthostatic hypotension, fainting, tachycardia, palpitations, chest pain; nausea, vomiting, constipation, or diarrhea; allergic reactions (skin rash, itching, urticaria, angioedema).
